Ekti Raat is a movie about two couples and their relationship. Sushabhan Dutta and his wife, Anita, were invited at Digbijay babu's place. Though Anita boarded the train but Shushabhan misses the train. There, in the railway station, he meets Santana, the wife of famous political leader Brajeshwar De who was also invited in Digbijay's babu place. As the destination is same, Shushabhan and Santana start their journey in the same car, expecting that they will meet their spouses at Digbijay's babu's house. But Anita returns home in search of her husband and came to know that her husband has gone to some other palce with another woman. Anita's mother Malina Debi was highly unsatisfied with the choice of life partner of her daughter and dislikes Shushabhan. Here she gets a chance to prove that Anita's marrige with Shushabhan was wrong and they all start a journey to catch Shushabhan red handed. On the other hand, Brajeshwar De came to know that his wife has spent a night in an hotel with a unknown person whom she met in the station. When Shushabhan and Santana reaches Digbijay babu's house, they come to know that Anita did not reach there. By calling at the home he comes to know that she is searching for him. On his way he met Brajeshwar babu and tries to convince him that nothing worng had happened. Finally all the charactes reach Digbijay babu's house to judge the truth and after a lot of confusion and drama, love wins over suspician. In this way the movie ends by ensuring that trust is the base of a successful married life.
