Sudhir Mishra’s passion for films came from his father, D.N. Mishra, a mathematician, who introduced the young Sudhir to world cinema. His father was a founder member of the Lucknow Film Society. Sudhir’s brother, Sudhanshu was a student at the Film Institute and it was he who taught Sudhir the art of film making. Later when Sudhir went to pursue his graduation in Delhi, he met Badal Sircar with whom he formed a theatre group called Workshop Theatre, and the two of them did a few plays together.

In 1984, Sudhir wrote the script for Saeed Akhtar Mirza’s Mohan Joshi Haazir Ho and in 1985 he scripted Khamosh for Vidhu Vinod Chopra. From here on, Sudhir graduated to directing his own films and he turned director in 1987. Till date he has directed 8 films and has written the script for 10 films. Sudhir has also made small appearances in many of his films, and some other films as well. In 2007 he played a major role in the film Traffic Signal. His forte as a writer and a director is parallel cinema. Sudhir’s directorial debut, Yeh Woh Manzil To Nahin (1987) won the National Film Award for Best First Film of a Director. After this came critically acclaimed films like Main Zinda Hoon (1988), Dharavi (1992), Is Raat Ki Subah Nahin (1996) and Chameli (2003), and his most accalimed film to date, the 2005 film, Hazaaron Khwaishein Aisi. This was followed by Dus Kahaniyan and Khoya Khoya Chand 2007. However both these films failed to create waves at the box office.

In 1987 Mishra won the Indira Gandhi Award for Best First Film of a Director for film Yeh Woh Manzil To Nahin. In 1989 he won the National Film Award for Best Film on Other Social Issues for Main Zinda Hoon. In 2006 he won the Filmfare Best Story Award for Hazaaron Khwaishein Aisi.
